Error: File in use when opening Android backup.ab

Using Kindle for Android 4.16.0.75 on my Lenovo Tab 4 8 tablet, I believe I've retrieved a valid backup.ab file, about 3 MB in size.
Does 3MB sound reasonable?

In Calibre DeDRM 10.0.3 customization, I select the Kindle for Android backup file and click open.

Then I get the following error message:

I eventually managed to run androidkindlekey.py by itself, but ended up with pretty much the same "used by another process" error.
